How

Unpack

Remove the bottom screw from the carrying case with the supplied screwdriver. Keep this screw

the

receptacle inside the case.

2. Shift the grayish-tan knob along the groove

the earring handle into the square opening tounlock

and

open the case.

3. Bring out the microscope from the case and remove the packing and covers.

Set up the microscope with the equipment such

objectives. eyepieces. mirror,etc.

5. The removed screw and packing may be rc·used for transportation. Tighten the screw when transporting.

IV Construction

Body

tube

Body tubes can

classified as follows

form and function.

Fixed tube

Vertical tube

Monocular tube

Adjustable

tube

Fixed

tube

Binocular

tube

(Trinocular) Adjustable

tube

The fixed

body

tube has a

constant

length,

that

is, the unchangeable distance between the eyepiece and

the

objective. In

Olympus

biological microscopes

the

tube

length is fixed

160

millimeters.

it will

2. Revolving nosepiece

customary for the objectives to

mounted

on the nosepiece in 3manner in which higher powers can

obtained when

the

nosepiece

turned clockwise with the right hand while looking through

the

eyepiece.

3. Stage

Square fixed stage. Mechanical stage can be filled

HS,

HSB

and HSC.

4. Focusing mechanism

The coarse adjustment mechanism consists

arack and apin-

ion by which the

body

tube

racked up and down. Models HS,

HSB

and

HSC have double-lever fine adjustment mechanism.

Stopper

Body tube stopper

This stopper

used for preventing the body tube from sliding

down farther

than

apre-set point. If the

stopper

fastened

tightly when the

body

tube has been adjusted

adesired level,

the tube can be racked

and down only above the pre-set level

but never come down below it. 'This arrangement prevents a

damaging collision between the objective

and

VHow to

Use

the Microscope

1. IUumination

What

most important

amicroscopic work

illumination, because the light source and condenser greatly

influence the resolving power

the microscope lenses.

For

microscope illumination, natural daylight

artificial light source is used. Normally

is sufficient

use

natural light coming through awindow. However, in some cases such as photomicrography, aspecially designed

microscope illuminator

used.

When

daylight

used

Avoid the direct rays

the sun.

most desirable that

the

microscope

placed

awindow facing north.

Usually

the

plane mirror

used but the concave side

used when annoying renections

the window frame

trees come into

the

field

view.

When

an illuminator

used

Alow-voltage bulb

used for microscope illumination.

Olympus puts on the market aspecially designed micro-

scope illuminator LSD

(u!:ing

a6V, SA transformer,

optical accessory). When this illumination apparatus

used,

the

beam

light must be adjusted by adjusting the

position

the bulb and

the

plane side

the

mirror should

used. The accessory daylight

mter

(cobalt color)

shoHld be inserted into

the

filter holder.

Centering

When amicroscope illuminator

used, adjust it

that

the

axis

the illuminating system falls in

the

center

the mirror. Next, rack

the

body tube up and down

that

it may be roughly in aposition in which

the

microscope

used for observing

the

specimen.

Remove

the

eyepiece and look through the eyepiece sleeve, and you will see

the

back lens

the

objective.

Adjust

the

mirror

that

the

back lens

evenly illuminated.

Similar procedures must

adopted when daylight

used.

Slide

glass

and cover

glass

microscopes are designed

use the slide glass and cover glass

the following thicknesses.

Slide glass: 0.8 -1.0mm

Cover glass: 0.16 -0.18mm

Use slide and cover glasses that satisfy these requirements.

2. Focusing

Afte~

completion

the centering

the

illuminating system, place the specimen

the stage and focus the

microscope on the specimen.

you move the body tube up

and

down while looking through the eyepiece. you

may

jam

the objective

down

the specimen, and you could

quite

easily crack the specimen and the front

lens as well.

First,

use

alow·

power

objective,

and

operate

the

coarse

adjustment

knob

to lower

the

body

tube

until

the

tip

the

objective

comes

the

specimen.

All

this

must

done

while

looking

the

microscope

from

the

side.

Next,

look

into

the

eyepiece

and

body

ext,

turn the nosepiece

magnification. Usc the fine

adjustment

knob

to get the specimen sharply in focus_

Move

the specimen by hand

operate the mechanical stage to bring the target area

the specimen into the center

the field

view.

3. Change

magnification

The magnifying power

the microscope

changed either

changing the eyepiece or by changing the

objective.

Change

the eyepiece

Eyepieces with different magnifications arc inserted into the eyepiece sleeve to change the magnifying power

the microscope. In this case, the microscope remains roughly

focus but further focusing

required to

obtain

asharply focused image.

2) Change

the Objective

When the objective

changed to change the magnification

the microscope, you first focus the microscope

with alow-power Objective

and

then turn the nosepiece to change to ahigher-power objective.

Model

you need only operate the fine adjustment

knob

obtain an accurate focus.

4. Resolving Power

The

resolving

power

amicroscope

depends

solely on

that

the

objective

and

has

nothing

with

the

eyepiece. The only function

the eyepiece

magnify the image resolved by the objective. This means that

matter

how

much the final magnification may

increased by using ahigh-power eyepiece, you will never

see what is not shown

the image that

formed

the objective. For example, you get the same magnifiC'dtion

800X

using a

40X

objective and a20X eyepiece,

and

also by using aIOOX objective and a

eyepiecc.

However. the latter combination has aresolving power twice higher and show more details than the former

combination.
